The Royal Randwick crowd cheered on as legendary jockey Glen Boss guided the colt home ahead of Santa Ana Lane in second and Trekking in third.
Trailing towards the end of the field at the last turn, Yes Yes Yes showed an excellent turn of foot to claim Australia's richest turf race.
Trainer Chris Waller said it was an exceptional performance by the three-year-old.
"It was a great win, especially when there is some much at risk," Waller told Channel 7.
"We basically had to go to all of the owners, Coolmore and the original owners, and tell them that you think this horse can win an Everest.
"Then you've got to go to the slot holders and tell them that we think this is the horse you should choose.
"It was a lot of weight on our shoulders and to tell you the truth, it's an amazing feeling."
An emotional Waller said his experience with former champion Winx had helped prepare him for Saturday's feature event.
"We're we've been with Winx, where she took us has helped set us up for days like this," he said.
"I think we took it in our stride this week and the reason I'm emotional is because of the journey she took us on."
Waller said he was confident The Everest would continue to grow into the future.
"Talking to people during the week, they know the runners in The Everest, they can relate to every single horse in The Everest," he said.
"Half of them might be back next year, might be more, because these are the best sprinters in Australia.
"That's what people want to see, they want to see the best sprinters racing two, three, four weeks prior.
"It's not just about one race and that's why I think The Everest will really go ahead."
Earlier in the day, the region's other runners shared mixed results at Royal Randwick.
Sue Grills' Burning Crown was scratched from The Highway Handicap, while Cody Morgan's Screen Shot ran third.
In The Kosciuszko, Mel O'Gorman's Suncraze could not repeat its heroics from last year, finishing in 13th position.
