The hosts, who had a bye in the first round due to being runners up against Orange's Kinross Walaroi School in last season's Shield grand final, lost the toss and were sent in to bat.
In three and a quarter hours at the crease opener Toby Smith knocked up 120 including 17 fours, before being caught by Hamish McFie in the 45th over. Captain Cotter Litchfield racked up 39 including a six and Zac Clarke knocked two over the fence before being trapped lbw for 37 by Lewis Koppel, who recorded the best bowling figures for Lindisfarne of 4-32.
TAS finished the innings with 7-257 after 50 overs.
Lindisfarne had a shaky start at the crease, with Smith catching Noah Chapman behind for a duck, and in quick succession Matt McPhee fell for two, the visitors at 2-9.
Playing his first Douglas Shield match for TAS, Zac Clarke was best of the bowlers for TAS with 3-45 and then Litchfield with 3-36.
For Lindisfarne Fletcher Koppel top scored with 50, caught by Andrew Kirk off Litchfield.
The visitors were 8-160 when thick smoke from bushfires rolled in from the east threatening an early finish to the match, but not before quick work from Litchfield bowled McFie for nine and in the next over Lachlan Galbraith caught Ethan Sekac for a duck, leaving Lindisfarne all out for 181.
"After we scalped two quick wickets Lindisfarne settled in and had a reasonable run rate, we just kept on top of the wickets," Litchfield said.
"For us, Toby Smith's innings was all class and everyone batted well down the line which kept the momentum up.
"It was a great result for us given it was the first Douglas Shield hit out for more than half our team, and the debut for the Firsts for Tom Younghusband and Louis Cannington."
The Douglas Shield is a knockout competition run by the NSW Country and City Schools Cricket Association which comprises all Independent Secondary Schools in NSW other than the Sydney GPS and CAS schools.
