A motorist reported the motorcyclist had crashed on the side of the Oxley Highway, about 7km east of Bendemeer.
The rider, a woman believed to be aged in her 60s suffered critical injuries and died at the scene.
Emergency services were called and are trying to piece together the moments leading up to the crash.
Investigators are calling for any witnesses or anyone who might have seen the rider along the Oxley Highway after 1pm on Wednesday.
Police remained at the scene late on Wednesday, with part of the road closed to traffic. A report will now be prepared for the coroner.
Wednesday afternoon
One lane of the Oxley Highway has re-opened following an earlier fatal crash between Bendemeer and Walcha Road.
Stop/slow traffic control is in place allowing motorists to travel through the area one direction at a time.
Police are still at the scene with one lane expected to remain closed for several hours.
Motorists are advised top allow extra travel time.

ONE person has died in a motorbike crash north of Tamworth on Wednesday.
Emergency services are at the scene at Bendemeer, after the accident on the Oxley Highway, about 7km east of the New England Highway intersection, towards Walcha Road.
A spokesperson for Ambulance NSW said one unit was deployed to the scene after a triple zero call shortly after 1.30pm.
The rider died at the scene.
Police have shut the Oxley Highway, with diversions in place via Thunderbolts Way and the New England Highway.

The Oxley Highway has been closed at the intersection of the New England Highway at Bendemeer and at the coroner of Derby and Fitzroy Street, Walcha.
The road is expected to remain closed for several hours.
A report will now be prepared for the coroner as police piece together the circumstances surrounding the crash.
